This time, Mario must save Princess Daisy, rather than Princess Toadstool, from her kidnapper Tatanga, a mysterious spaceman. The game follows largely the same formula of its aforementioned predecessor, with Mario defeating enemies by jumping on them and collecting coins and power ups by hitting floating blocks, with each world leading up to a boss fight which is defeated using by collapsing the floor. A unique feature of this installment is two scrolling shoot-'em-up levels, the first of which Mario pilots a submarine, and the second an aeroplane.

Dr. Mario is a falling block tile-matching video game, in which Mario assumes the role of a doctor, dropping two-colored medical capsules into a medicine bottle representing the playing field. This area is populated by viruses of three colors: red, yellow, and blue. In a manner and style considered similar to Tetris, the player manipulates each capsule as it falls, moving it left or right and rotating it such that it is positioned alongside the viruses and any existing capsules. When four or more capsule halves or viruses of matching color are aligned in vertical or horizontal configurations, they are removed from play. The main objective is to complete levels, which is accomplished by eliminating all viruses from the playing field. A game over occurs if capsules fill up the playing field in a way that obstructs the bottle's narrow neck.

Manhole was remade with an updated "Modern" version for Game & Watch Gallery. Toad, Donkey Kong Jr., and Mario are the pedestrians in this version, walking at different speeds, while Yoshi is the character the player controls. In this version, four manhole covers can stay up at once, unlike in the original. The manhole covers can drop from characters stepping on them, causing the next pedestrian to fall if Yoshi does not act quickly. Yoshi can also hold a manhole cover in place to keep it from falling. He holds up the upper manhole covers with his head and the lower ones with his tongue. At 200, 500, and 700 points, a Chubby comes out of the ground and throws a heart for Yoshi to catch to remove a miss. This version also features music, with the tempo changing depending on the speed of the game.

Every game in the main series has a standard Party Mode in which up to four players play through a board, trying to collect as many stars as possible. In every turn, each player rolls a die and progresses on the board, which usually has branching paths. Coins are primarily earned by performing well in a minigame played at the end of each turn. On most boards, players earn stars by reaching a star space and purchasing a star for a certain amount of coins. The star space appears randomly on one of several pre-determined locations and moves every time a star is purchased, usually occupying a blue space. Every Mario Party contains at least 50 to almost 110 minigames with a few different types. Four-player games are a free-for-all in which players compete individually. In 2-on-2 and 1-on-3 minigames, players compete as two groups, cooperating to win, even though they are still competing individually in the main game. Some minigames in Mario Party are 4-player co-op, even though it doesn't say it. In most situations, winners earn ten coins each.

Luigi and his opponents start with five hit points each, and they reset to five each time one of them gets pushed back. As the player advances in the game, the enemies get more difficult to defeat. Getting one KO earns a coin, getting five earns a Yoshi Coin, getting ten earns a trophy, and getting 50 earns a Championship Belt. It is Game Over if Luigi gets KO'd.

The Modern version has a few differences. First, Mario replaces Stanley (just like how an Orange Yoshi replaces him for the Modern mode of Greenhouse in Game & Watch Gallery 3). Mario and Donkey Kong also have bubble shooters instead of insecticide sprays. Plus, the setting is a Ghost House. Lastly, the bees have been replaced by a fireball and a Boo. The fireball remains in place until pushed away by the bubbles. The Boo attempts to sneak up on either Mario or Donkey Kong when one of them is busy refilling his bubble shooter. When the fireball or Boo reaches Mario's or Donkey Kong's side, whoever receives it freaks out. Unlike in the Classic version, music is featured.

The Modern version of Fire Attack involves Wario, who is trying to protect a giant golden statue of himself from a bunch of Bob-ombs and Bullet Bills. During his defense, however, Wario has to be careful not to hit any hens or apples, as they award him five and seven bonus points, respectively. The player is not penalized for hitting a hen or apple, but the player does get a miss if a Bob-omb or Bullet Bill manages to destroy parts of Wario's fort. However, Wario can remove a miss when he gets 200, 500, and 700 points by catching a heart coming from a Bill Blaster. It is Game Over if Wario's statue gets destroyed. This version also features music, with the tempo changing depending on the speed of the game.
